What is it about?
The elevating resistance of insect pests to conventional insecticides has heightened the urgency to implement novel chemical scaffolds that offer enhanced efficacy, selectivity, and safety profiles. Among these, isoxazoline analogs have evidently emerged as a prospective class of insecticides, demonstrating broad-spectrum behavior against various agricultural and urban pests. This review aims to integrate a thorough assessment of the synthetic, structural, and insecticidal advancements of isoxazolines and isoazoles from the 2016 to 2026.

Why is it important?
Isoxazoline and isoxazoles have emerged as promising insecticides in urban and agricultural settings, attributable to their novel mechanisms of action and broad-spectrum efficacy. Significant advancements have been instituted in the eco-friendly and efficacious synthesis of isoxazoline and isoxazole-based molecules. The structure-activity relationship (SAR) studies have identified imperative modifications that enhance the insecticidal potency and compound selectivity. Commercial isoxazoline and isoxazole insecticides have illustrated outstanding effectiveness against resistant pest populations in both urban and agricultural environments.
Perspectives
Structure-activity relationship studies have identified key modifications that improve insecticidal potency and selectivity while emphasizing the possibility of hybrid molecules to counteract resistance mechanisms. In addition to their high efficacy, isoxazoline- and isoxazole-based insecticides generally illustrate favorable toxicological and environmental profiles, posing reduced risks to humans and non-target organisms compared to many traditional insecticides. The review further discusses the mechanisms of action of isoxazolines, their incorporation into integrated pest management (IPM) strategies, and their commercial applications in urban and agricultural pest control. Lastly, this article addresses current challenges and future research directions, underscoring the necessity for ongoing innovation and safety assessments to ensure the sustainable use of isoxazolines and isoxazoles in modern agriculture.
