What is it about?
We try to explain firstly how the concept of the initiator of DNA replication(DnaA) in E.coli is conceived and then how DnaA was isolated .The in vitro studies on DnaA and oriC brought about the explanation on how DnaA opens oriC,licensing of initiation. We try to reevaluate these in vitro studies compared to those in vivo which produced the concept of Initiation Mass. Introducing the nucleotypic effect,the possibility is dicussed about the Initiation mass as the consequence of initiation rather than a launching element.Another question of how desequestration of hemimethylated oriC is examined taken into account the cellular metabolisms.
