What is it about?

The paper titled “A Holistic Review of Oncological Drug Targets and Trajectories of Resistance in Cancer Therapy” is a review article about modern cancer treatments and why cancers often become resistant to them. It discusses both established targets (such as HER2, hormone receptors, and angiogenesis pathways) and newer precision-medicine targets like KRAS G12C, CLDN18.2, and TROP2. The review also explains mechanisms of drug resistance, challenges in targeted therapy, and how technologies such as genomics, omics, and artificial intelligence may improve future cancer drug discovery and personalized treatment.

Why is it important?

This paper is important because it provides a comprehensive overview of both current and emerging cancer drug targets while also explaining how and why tumors develop resistance to therapy. By combining knowledge on targeted treatments, resistance mechanisms, and new technologies such as genomics and AI, it helps researchers and clinicians better understand the challenges of precision oncology and guides the development of more effective, personalized cancer therapies.

Perspectives

From my perspective, this publication highlights the urgent need to better understand both oncological drug targets and the mechanisms behind therapeutic resistance. By integrating insights from targeted therapy, molecular oncology, and emerging technologies such as genomics and AI, I aimed to provide a broad yet clinically relevant overview that may support the development of more precise and durable cancer treatments in the future. Researchers can benefit from this work by using it as a consolidated reference for current and emerging therapeutic targets, resistance pathways, and future research directions in precision oncology.
