What is it about?

Back in 2015, we described one pvl positive-VISA isolate of the USA1100 (ST30) lineage. In this paper we performed the genome sequence of this isolate, to better understand the genomic features that could be related to the establishment of this lineage in our country, and also in Latin America. The sequence showed that the core genome of our isolate was pretty similar to the genome of other S. aureus. However, it contained differences in the accessory genomes such as two plasmids, six genomic Islands, and four prophages. Interestingly, besides the pvl gene, we showed the presence of a prophage containing two new pvl genes. Besides that, our isolate harbored genes for serine proteases, the enterotoxin gene cluster, the immune invasion cluster, spermine/spermidine acetyltransferase gene, previously detected only among USA300 isolates, a lineage that is epidemic in the USA and other countries, but not in Brazil. Moreover, we observed that the isolate sequenced produced 5 times more PVL messenger RNA when compared to a USA300 reference strain. Plus, regarding the VISA phenotype, we detected mutations at vraS, tcaA and B genes.

Why is it important?

Staphylococcus aureus is an important cause of infections worldwide, and the antimicrobial resistance still a serious public health problem, particularly the resistance to b-lactams and the intermediate resistance to vancomycin. Besides, it presents several virulence genes, such as Panton-Valentine Leukocidin (PVL). In Brazil, pvl positive S. aureus related to the USA1100 (ST30) lineage is disseminated in both community and hospitals. In this work, we were able to do the first description and gene sequencing of a pvl positive S. aureus isolate, presenting the VISA phenotype, associated to the USA1100 lineage. We were able show some virulence features that may contribute to the dissemination success of this particular lineage in Latin America countries including Brazil.

Perspectives

With this work we hope that we can help other groups to further investigate what makes some S. aureus lineages more adapted to certain countries than others, as is the case of USA300 , in USA, and USA1100 in Latin America.
