What is it about?
We analyzed the SOS images of formalin-fixed paraffin-embedded skin sections from elderly, young, diabetic, and nondiabetic subjects, as well as chronic and acute wounds. SAM provided high-resolution histology similar to LM and revealed characteristic SOS alteration following pepsin treatment.

Why is it important?
The sound speeds through skins reflect the stiffness of the lesion, which are statistically comparable among various states of the skin lesions. We can estimate the age, the severity of DM, and the progress of the wound.
Perspectives
I hope the method using scanning acoustic microscope will become familiar to other scientists. This microscope provide not only fine histology but also mechanical properties of the tissue. Its advantage over light microscopy are as follows: 1.No staining is necessary. 2.It takes a few minutes to obtain histology images. 3.Obtained images are so originally digital that statistical analysis is easy. 4.Repeated observations after operating the section are possible such as protease digestion. Because higher resolution is available as sound frequency increases, observation of cytoplasmic organella and intranuclear structure is possible experimentally.
