What is it about?

We designed a next-generation sequencing test covering the complete HBB gene and validated it in a Pashtun family affected by beta-thalassaemia. The affected family member carried a pathogenic insertion in both copies of the gene, while unaffected relatives carried one copy. The approach also detected an additional synonymous variant.

Why is it important?

Traditional targeted tests can miss uncommon or family-specific variants. Sequencing the full gene can provide a more comprehensive diagnosis, support carrier detection and strengthen genetic counselling in populations where consanguineous marriage and beta-thalassaemia are common.

Perspectives

From my perspective, this work matters because Traditional targeted tests can miss uncommon or family-specific variants. I hope the findings support further research and informed, evidence-based decisions.
