What is it about?
We engineered the repDE ribosome-binding site of the pTRKH3 plasmid by site-directed mutagenesis to increase plasmid copy number in Lactococcus lactis LMG19460 cells. 215 copies of plasmid per chromosome were achieved, a 3.5-fold increase when compared to the nonmodified plasmid.

Why is it important?
The generally recognized as safe Lactococcus lactis could constitute a safer alternative for plasmid production. A key requirement for the establishment of such a cost-effective manufacturing is the availability of high-copy number plasmids.
