What is it about?
This preprint examines whether a phosphate buffer would help ameliorate large decrease in pH in bacterial culture broth supplemented with glucose, and help improve biomass formation. In addition, the work also examine whether supplementation of nitrogen sources is necessary to improve biomass formation in LB medium.

Why is it important?
The work shows that a large capacity phosphate buffer is capable of reducing large drop in pH arising from the utilization of supplemented glucose in LB medium by E. coli, where a more conducive pH environment helps improve biomass formation. In addition, LB medium was found to be not deficient in nitrogen sources, where supplementation of nitrogen sources did not improve growth.
Perspectives
This preprint provides important data demonstrating the need for adding a phosphate buffer system in growth medium for taking advantage of the growth potential inherent in supplementing the medium with glucose.
