Tadalafil is a drug that is used to treat male erectile dysfunction. In this paper we prepared some pharmaceutical molecules that can work more potently than tadalafil and even more selective. This can help famous muscle pain associated with tadalafil.

The paper shows an easy, accessible method for synthesis of the nucleus of tadalafil. It highlights how to seperate isomers produced and suggest some chemical modifications to improve potency and selectivity.

PDE5 is gaining more attention not only for MED but also for pulmonary hypertension, cancer and Alzheimer. This work is a foundation for those who wants to develop PDE5 inhibitors for new indications.
