What is it about?
This paper is about the state-of-the-art of IDH inhibitors considering a medicinal chemistry point of view. Besides that, we discuss the rational approach that was employed for the discovery of IDH inhibitors and the molecular profile of the main inhibitors described in the literature, considering the potential that can be explored in the early future.

Why is it important?
This review highlights, differently from most papers about this theme, the historical landscape that allows us to comprehend how some genetic and biochemical aspects resulted in the discovery of this target. Besides that, we propose some new strategies that can be employed for the discovery of new IDH inhibitors.
Perspectives
We hope that this paper can be useful for those that desire to understand the molecular basis of actions of IDH inhibitors, with the aim to contribute for discovery of new IDH inhibitors.
