What is it about?

The diversity of stain fungi is important if wood is inhabited with various fungi, and the discoloration mechanism will be better understood. MiSeq amplicon high-throughput sequencing (Illumina®) is able to detect species richness (the number of species within a community) and species evenness (the sizes of species populations within a community).

Why is it important?

This study demonstrated the first time that the stain fungal diversity in discolored wood can be detected by the MiSeq amplicon high-throughput method, focusing on the nuclear ribosomal internal transcribed spacer-1 (ITS1). Several fungi participate in the discoloration, with the predominant fungi (90%) being Ascomycota.

Perspectives

It is important that the MiSeq approach also delivers data about the composition of the fungal community, as the universal seven fungal species were identified. The method presented is more rapid and economic than the T-RFLP approach. Moreover, cloning is not required.
