A holographic microscopy algorithm is proposed enabling to deal with high numerical aperture holograms. Hologram reconstruction kernel performs under afocal conditions, which allows keeping the full NA over wide defocus range.

Why is it important?
We have proposed an holographic microscopy reconstruction method compatible with high numerical aperture microscope objective MO. The key idea is to apply in the plane of the camera a phase mask (RM+NL) that transforms the optical elements of the holographic setup into an afocal device. This mask makes also the standard phase corrections: phase curvature induced by the microscope objective, phase curvature of the reference, and off axis phase.
Perspectives
We proposed a two step reconstruction method operating with high numerical aperture holograms (up to NA = 1.4).The benefits of the methods have been demonstrated through the reconstruction of holograms recorded with a ×60 NA = 1.4 oil immersion MO, proving the ability to keep almost full resolution in a [−50μm;50μm] defocus range.
