Nebivolol is a highly beta1-selective beta-blocker with additional NO-mediated vasodilatory effects, and it is well known that continuous long-term treatment with nitrates may cause nitrate tolerance

Why is it important?
In our randomised, double-blind, placebo-controlled, cross-over study oral administration of 5mg Nebivolol daily for eight days did not decrease the vasodilatory effect of nitrates
Perspectives
Therefore, we conlcude that there is no evidence of nitrate tolerance caused by long-term administration of Nebivolol. In contrary, in our study chronic intake of Nebivolol rather increased than decreased NO-mediated vasodilating effects
