Detection by Flow Cytometry of anti- DNA autoantibodies and immune complexes

What is it about?

A new method for the detection by flow cytometry of anti- DNA antibodies and of circulating immune complexes containing endogenous DNA is described.

Why is it important?

Autoantibodies to double-stranded DNA are the serological hallmark of systemic lupus erythematosus (SLE). When anti-DNA antibodies are neutralized by endogenous DNA and can no longer be detected by routine tests, the serologic diagnosis and the follow-up of relapses in patients with SLE is compromised. To overcome this obstacle, we propose an accessible solution: the detection of circulating DNA immune complexes by flow cytometry.

The results of routine tests for the detection of anti-DNA autoantibodies do not correlate well. This could be due to the fact that lupus serum samples contain both anti-DNA autoantibodies, endogenous DNA and immune complexes. In the near future the tests should be designed and the results interpreted by integrating this data.
