What is it about?
We examined the prostaglandin receptor subtype involved in DOMS. The mechanical withdrawal threshold of the exercised muscle was measured before and after LC in rats administered prostaglandin E2 receptor (EP) antagonists before LC, or in wild-type (WT), EP2 knockout (EP2−/−), and IP knockout (IP−/−) mice.

Why is it important?
Our findings suggest that EP2 contributes to generating mechanical hyperalgesia through positive feedback upregulation of COX-2 expression in muscle after LC.
Perspectives
The present experiment has shown EP2 plays a major role in the early stage of generation of mechanical hyperalgesia after LC. As involvement of PGE2 has been indicated also in humans,[23] EP2 may play an important role also in human DOMS. Importance of exercise for fitness and lively aging is year by year more strongly recognized, and prevention from any painful condition related to exercise is desirable for promoting exercise. Studies linking our results and human studies such as prevention of DOMS by preconditioning of exercise[40] will bring better understanding of mechanism of DOMS.
