We benchmarked cryoSPARC helical reconstruction on deposited and in-house amyloid cryo-EM datasets, tested current capabilities, identified key limitations, and present comprehensive user guidelines for optimized amyloid processing workflows.

Why is it important?

Effective software development relies on community involvement and feedback. To expand cryoSPARC into a truly universal processing pipeline capable of handling both single-particle analysis and helical reconstruction, comprehensive guidelines for reconstructing amyloids need to be established and formalized.

Perspectives

Our goal is to shift the focus of amyloid cryo-EM from computationally intensive processing challenges towards addressing fundamental biological questions that enhance our capacity for treatment discovery.
