What is it about?

.........The importance of powder diffraction to determine the new solid phases.........Cocrystals of acemetacin drug were prepared through melt crystallization and their X-ray crystal structures determined by high-resolution powder X-ray diffraction. The powerful technique of structure determination from powder data (SDPD) provided details of molecular packing and hydrogen bonding in pharmaceutical cocrystals of acemetacin. Among all nicotinamide cocrystal resulted in anhydrate and hydrate forms. We also provided the details explanations of the all related studies in the direction of pharma cocrystals.

Why is it important?

All the characterizations suggest nicotinamide cocrystal new solid phases may be polymoprhs. As the drug and cocrystals prone to hydrate during solvent crystallizations, we could not get single crystals. So the confirmation of the hydrate and anhydrate nicotinamide cocrystal happens only with powder data. This study opens a great door and confirms the importance of powder data in pharma cocrystals area. Apart from powder data, the conformational study reveals the great effect of the new solid guest to result from target conformation.

Perspectives

It will be great interest to look the physicochemical and pharmacokinetic studies further. finding the best pioneer one from the reported library will be the alternative to the parent drug.
