What is it about?
This study used hydrophilic interaction liquid chromatography (ZIC-HILIC) to examine the chromatographic behavior of tramadol hydrochloride. For the chromatography inquires, two lab made HILIC column with distinct spacer lengths between the ions sites collections ZIC-2 and ZIC-3 were utilized. The research specifically examines the effects of different spacer length on tramadol retention patterns. The primary objective of the experiment was to investigate a number of separated factors; including pH, buffer amount and acetonitrile quantity in the solution. In the investigation acetonitrile and acetate buffer were combined in 90:10(v\v) ratio. With pH 5, the buffer ionic strength was 25mM. 0.5, l\min flow rate was employed for the eluted using an isocratic method. Utilizing UV detector calibrated to operate at certain wavelength 273nm, the molecules was located. Based on the results, recognizing tramadol in drug can be done directly using ZIC-HILIC mode. The HILIC method employed in the research yielded high accuracy (RSD<1%) and significant linear connection between the tramadol concentration and the caliberation. A range of 0.1-6.3 μg\mL was found for the tramadol content. For tramadol in ZIC- and ZIC-3, respectively, the HILIC methods indeacted good LOD of 0.080 and 0.050 μg\mL and LOQ varied from 0.242 to 0.151 μg\mL. In ZIC-2 and ZIC-3, the main tramadol recovery percentage were (98.80%-100.3%) and (98.6%-101.3%), respectively.

Why is it important?
Tramadol is a commonly prescribed opioid, but its misuse and improper dosage can have serious consequences. Existing methods for analyzing tramadol are often time-consuming or lack sensitivity. This study introduces a simple, cost-effective, and highly accurate approach for detecting tramadol in drugs, ensuring better quality control in the pharmaceutical industry. The method’s low detection limit and high accuracy make it useful for both regulatory agencies and pharmaceutical companies.
Perspectives
Pharmaceutical Industry: The proposed method offers a reliable way to test tramadol content in medications, ensuring safety and efficacy. Regulatory Bodies: Authorities can use this method for drug quality control and to detect potential counterfeit medications. Healthcare Providers: Doctors and pharmacists can rely on improved drug testing to minimize incorrect dosages and ensure patient safety.
