What is it about?

A thermostable Cas9 from Acidothermus cellulolyticus (AceCas9) was characterized in vitro and demonstrated its activity under in vitro and in vivo conditions. Interestingly, through biochemical assays, activity of AceCas9 shown to be DNA topology-dependent. This publication illustrated the new Cas9 system is optimal at elevated temperature and with longer guide during DNA targeting. With the novel PAM motif 5'-NNNCC-3', this work further expands the molecular toolbox of Cas9 that are naturally found in bacteria.
