What is it about?
We developed a TaqMan real-time PCR test for the MPL W515L and W515K mutations found in some myeloproliferative neoplasms. The assay detected mutant levels down to about 5%. Among 128 patients, two carried W515L and one carried W515K, showing that the method could detect uncommon low-level variants.

Why is it important?
MPL mutations help diagnose myeloproliferative neoplasms but may be missed when the mutant fraction is small. A faster, less expensive and more sensitive test can improve access to molecular diagnosis and support measurement of mutation burden.
Perspectives
From my perspective, this work matters because MPL mutations help diagnose myeloproliferative neoplasms but may be missed when the mutant fraction is small. I hope the findings support further research and informed, evidence-based decisions.
