In this review, we summarized the current findings on neuroimmune Sema4A and Sema4D molecules in chronic inflammation underlying many diseases and discussed their positive or negative impacts on the implicated molecular and cellular processes.

This review will help with a further development of Sema4-based strategy as a novel therapeutic approach for chronic inflammatory diseases such as asthma, COPD, IBD, SLE, MS, and cancer.

Sema4A and Sema4D as critical regulatory molecules in chronic inflammation with multiple distinct functions including, but not limited to, immune cell costimulation, regulation of cell morphology, adhesion, proliferation, stability, and migration.
