This work is about MS analysis of protein O-glycosylation. Here, deglycosylated/alkylaminylated tryptic peptides were submitted to Q-TOF MS/MS using CID, which allowed the identification of modified AA residues.

O-glycosylation represents an important posttranslational modification (PTM) of proteins, and is particularly relevant for glycoscience.
Perspectives
This method represents a simple and elegant tool for identification of O-glycosylation sites in glycoproteins. However, the deglycosylation/alkylaminylation reaction is problematic when densely glycosylated regions of glycoproteins are analyzed.
