Advantage of parallel measurement of antibody binding and complement activation on antigen chips

What is it about?

We measured both the binding of antibodies and of complement components to antigens on microarrays (chips) following immunization of mice using different forms of the same hapten antigen and different routes and adjuvants for immunization. Compared to ELISA tests where distinct murine IgG isotypes had to be measured to tell the difference between immunizations the "two-dimensional" chip measurements were sufficient to identify and differentiate groups receiving different treatments.

Why is it important?

The complement system is an important effector arm of antibody-mediated immunity. By measuring antigen specific antibody binding and complement activation together we obtain a deeper insight into the hosts reactivity against a given set of antigens.
