What is it about?

NMR is a well established technique utilized by chemists the world over to elucidate chemical structures. The majority of chemists at this point are used to 1H and 13C NMR and a small array of 2D experiments. Many are not aware of how 15N NMR, despite the nucleus being a very rare spin, is now accessible with modern NMR techniques, especially via indirect techniques. This chapter discusses various aspects of applying experimental AND predictive N15NMR to alkaloid structure elucidation

Perspectives

The utilization of 15N NMR chemical shift data in structure elucidation studies is now 20 years old. Although direct detection of 15N data remains a significant challenge due to both the low natural abundance and gyromagnetic ratio of 15N, in contrast, these data can be readily had by inverse detected experiments that access 15N chemical shift data via the high abundance and sensitivity of proton using either 1JNH or nJNH coupling pathways to detect the 15N. Alkaloids, by definition, all contain 15N in their structures. The utilization of 15N chemical shift data is pivotal to the elucidaiton of alkaloid structures.
