What is it about?
Three screening strategies based on 2D chromatography were reviewed, including traditional separation-based screening, bioactivity-guided screening and affinity chromatography-based screening. Meanwhile, in order to cover these aspects, selections of different separation mechanisms and modes were also presented.

Why is it important?
Screening of natural bioactive components using 2D chromatography helps separation and analysis of complex samples with greater targeting and relevance, which is very important for development of innovative drug leads.
Perspectives
Emergence of analytical methods based on comprehensive 2D chromatography has become a distinct trend in natural product research. It is a strategy that focuses on the global characterization of the sample and allows heuristic understanding of the interrelationships between different components in complex samples. Surprisingly, despite the advent of comprehensive 2D chromatography, its usage has not been fully exploited yet in screening of bioactive natural product. There are still many limitations to its practical diffusion, such as high cost of instrumentation, complex data processing systems and lacking of corresponding methodological validation indicators. However, if these challenges can be solved, the method of bioactivity screening based on comprehensive 2D chromatography will certainly become a powerful tool to advance natural product research and gain deeper knowledge in the field.
