How a herbal formula reshapes gene regulation in a rat model of liver cancer

What is it about?

The environment surrounding regenerating liver cells can influence how liver cancer develops. One process involved is DNA methylation—the addition of chemical tags to DNA that can change gene activity without altering the underlying genetic sequence.

This study investigated whether Diwu Yanggan Capsule, a traditional Chinese herbal formulation, could modify DNA methylation in the liver regeneration microenvironment. Researchers divided 48 rats into three groups: a healthy control group, a liver cancer model group and a model group treated with Diwu Yanggan Capsule for 16 weeks. Compared with healthy rats, the liver cancer model showed altered methylation in the promoter regions of 2,422 genes. These genes were associated with processes including immune responses, steroid hormone production and drug metabolism.

Treatment with Diwu Yanggan Capsule altered the methylation status of 1,650 gene promoters compared with the untreated model. Many of these genes were involved in calcium and cAMP signalling and other pathways associated with cell growth, cell death and regulation of the tissue environment.

The researchers examined several candidate genes in greater detail. Diwu Yanggan increased methylation of the ADCK2 gene promoter while reducing its RNA and protein expression, suggesting one possible epigenetic pathway through which the formulation acts in this animal model.

Why is it important?

Abnormal DNA methylation can disrupt the liver’s normal regeneration processes and create conditions that support cancer development. Understanding whether these changes can be modified may reveal new targets for preventing or slowing liver cancer.

This study provides mechanistic evidence that Diwu Yanggan Capsule can influence a broad network of epigenetic signals in a rat model, with ADCK2 identified as a potential target for further investigation. It also connects a complex herbal formulation with measurable changes in gene regulation rather than describing its effects only at the tissue level.

However, the research was conducted in rats and did not demonstrate that Diwu Yanggan prevents or treats liver cancer in humans. Further studies are needed to establish how ADCK2 contributes to liver cancer, identify the active components responsible for the observed changes and determine whether the findings have clinical relevance.
